Semaglutide is a long-acting glucagon-like peptide-1 (GLP-1) receptor agonist approved for type 2 diabetes management and chronic weight management. Clinical trials demonstrate significant weight loss potential averaging 15–20% body weight reduction through appetite suppression, slowed gastric emptying, and enhanced glucose control. Acylated with a C18 fatty diacid via a γ-Glu spacer, semaglutide achieves a ~7-day half-life enabling once-weekly subcutaneous dosing. Mechanism of Action

Its C18 fatty diacid modification enables albumin binding, prolonging circulation time and allowing sustained receptor activation with once-weekly dosing.
